It is also worth mentioning that multiple components of the InsR pathway, including age-1, pdk-1, akt-2, and daf-16, are predicted to be targets of the let-7 family miRNAs. miR-71 Is Not Required for Arresting Seam Cell or M-Cell Divisions During L1 Diapause.
Games without play are contests or performances, or both – but not true games. Conversely, having abundant experience of respect and gratitude for fellow players can help us develop habits of moderation, perspective, self-control, and empathy when it comes to non-game frustrations. Rather, we should be – you guessed it – intelligently curating and sharing the more pro-social alternatives, and giving people context and vocabulary for their engagement with culture. Not by running “don’t play Monopoly” campaigns, any more than we run “don’t read mass-produced paint-by-numbers genre pulp that robotically replicates toxic social norms” or “don’t watch reality exploitation TV” campaigns. Our job is to intelligently curate and share the worlds of culture and information in service to our communities, helping people find context and vocabulary for their engagement with the wider world. And as we’ve discussed above, simply offering the kind of safe, supported opportunities to play that we’d want to offer anyway is enough to create that space for healing.
Given my views on play, you won’t be surprised to learn that I asked revery play the presenters about whether play experiences could qualify as the kinds of responsive, positive, engaged, and empowering interactions they described. I was attending professional development aimed at helping library staff support families from refugee backgrounds, and we were discussing the role that libraries and childcare staff (mostly the latter, based on the mix of the group) can play in helping often highly traumatised communities. A small increase in risk of harm, replicated many times on a massive scale, means harm is certainly occurring. As COVID-19 lockdowns unfolded around the world, and sales of games, puzzles, toys, and other playful artforms spiked, a friend asked me if I felt a sense of validation. And worst of all, a combination of the two, where other players essentially become supporting cast for a scripted victory that is neither earned, nor fun, nor remotely playful – a psychodrama rather than a game, toying with their victims rather than playing with them. Controlling bullies who insist everyone play their way, forcing people to make deals they don’t want and to keep playing even when they aren’t having fun.

This result suggests that the high expression of miR-71 during L1 diapause is induced or maintained by other signaling pathways. We asked whether the expression of miR-71 was regulated by DAF-16, which is required during L1 diapause for long-term survival (2). It is possible that other miRNAs, including those in the let-7 family, control developmental timing in other tissues during the recovery phase after L1 starvation. We speculate that the expression of heterochronic genes controlling the L2/L3 programs, including that of hbl-1 and lin-42, are increased during L1 diapause to arrest the developmental progression, and miR-71 is probably required to suppress these “excess” signals during the recovery phase (Fig. S5). As pointed out above, multiple miRNAs in addition to miR-71 and the let-7 family miRNAs have roles in L1 diapause, and they may regulate the expression of many diverse targets that may include, but are not limited to, factors involved in UNC-31–InsR-signaling activities. Although the complete removal of miRNA functions causes embryonic lethality or infertility in worms, a partial disruption of overall miRNA functions by mutating either ain-1 or ain-2 provides an effective way to investigate miRNA functions (16, 17).

Non-Unc stable transgenic lines were maintained, and the expression of GFP and mCherry were observed under a Zeiss Axiovision II microscope. Three days later, the number of worms that were L2 or older was recorded as number of survived worms (Ns), and the survival rate was calculated as Ns/Np, which is an estimation of survived worms in the whole population. MT12993 mir-71(n4115) worms were outcrossed with N2 for four generations before any test except the initial screen. A recent study showed that the expression of miR-71 was significantly increased relative to other miRNAs in starved L1 worms (15). However, miR-71 does not appear to regulate all postembryonic development during L1 diapause recovery. Unlike classical heterochronic miRNAs such as lin-4 and let-7, the role of miR-71 in vulval cell division is essential in animals recovering from starvation-induced L1 diapause, but not in animals hatched on plates with food.

Which leads me to add to my wholehearted endorsement of Gene’s call for libraries to make allowances for people to avoid play and games. The fact that family game night might, as Gene remarked, be a source of trauma is primarily an indictment of the state of those families – but given play’s links to health and the fact that games can heal and promote bonding and caring, it seems unwise to ignore the qualities of the games they are playing. It’s easy to take this as a joke – families fight over board games, oh well, It’s Only A Game – but if you dig even slightly into this, you find that some of these experiences are genuinely traumatic, as one would expect might be necessary to give someone a lifelong aversion to an entire medium. Monopoly is… as legendary for spawning bitter family fights and for dragging on interminably even though it’s obvious who’s going to win as it is for being widely played. The spending limit for a child account is set to 0 by default when the account is created by the family manager.
